It is essential for the proliferation and development of megakaryocytes, leading to an increase in circulating platelet levels in the body. TPO operates through the c-mpl receptor, playing a pivotal role in maintaining platelet numbers in the bloodstream. Notably, TPO from humans and mice is capable of cross-species interaction. The human TPO gene produces a 353 amino acid glycoprotein, inclusive of a 21 amino acid signal sequence, a 15 amino acid segment akin to the erythropoietin domain, and a significantly glycosylated 179 amino acid sequence at the C-terminal. The recombinant form of Human TPO, a fully active 174 amino acid polypeptide (18.6 kDa), incorporates the erythropoietin-like domain found in the full-length TPO protein, showcasing its bioactive potential.
